Nicotine is the additive compound in tobacco smoke. nicotine which affects our  brain and it results in dependency.

Smokers intake several carcinogenic pyrolytic products along with tobacco smoke that bind to DNA which result in genetic mutations. The tar compounds is tobacco smoke (polynuclear aromatic hydrocarbons ) like benzopyrene and acrolein can can toxicate the cell’s nuclear DNA, which may either kill the cell or cause a genetic mutation. If the mutation inhibits programmed cell death, the cell can survive to become a cancer cell.

Small amount of radioactive compounds  get into tobacco leaves from the mineral deposits in the soil -  lead-210 (210Pb) and polonium-210 (210Po) are the radio active compounds found in tobacco smoke . Lead 210 is a product of the decay of radium-226 and, in turn, its decay product, radon-222; lead 210 then decays to bismuth-210 and then to polonium 210, emitting beta particles in both steps. These radio active compounds entering into our body is capable of damaging the tissues of our body.
